Optogenetics has a lot of potential to become an effective neuromodulative therapy for clinical applications. Selecting the correct opsin is crucial have optimal optogenetic tool. With computational modeling, neuronal response current dynamics can be extensively and systematically tested. Unlike electrical stimulation where effect directly defined by applied field, in optogenetics indirect, depending on selected opsin's non-linear kinetics. continuous expansion possibilities, studies are difficult due need accurate model first. To this end, we propose double two-state as alternative conventional three four state Markov models used modeling. Furthermore, provide fitting procedure, which allows autonomous starting from vast parameter space. successfully fitted two distinctive opsins (ChR2(H134R) MerMAID). Both able represent experimental data with great accuracy were obtained within acceptable time frame. This absence differential equations enormous reduction cost result. The performance proposed fit ChR2(H134R) was tested, comparing neural regular spiking neuron non-instantaneous, (4SB), derived Williams et al. ( 2013 ).
